Whole-genome sequencing of EMS-mutagenized adult Drosophila to identify modifiers of EndoA function

Endophilin A (EndoA) is a critical regulator of synaptic autophagy and is functionally linked to other proteins implicated in synaptic maintenance and Parkinson s disease. To uncover additional genetic modifiers of EndoA, we performed an unbiased mutagenesis screen using ethyl methanesulfonate (EMS) in Drosophila melanogaster. Mutant lines were screened for their ability to rescue the electrophysiological (ERG) defects observed in endoA mutants. The sequenced samples represent lines that successfully rescued the endoA phenotype and were analyzed to identify candidate modifier genes. As a reference, an isogenized (iso) control line was included, which carries known cn bw mutations and serves as the background for EMS mutagenesis. The iso line is also optimized for the LIND assay used in neurodegeneration screening.
