The liver participates in multiple functions, responsible for metabolic activities, nutrient storage and detoxification, that are mostly performed by hepatocytes and critical for maintaining physiological homeostasis. In addition, the liver engages various mechanisms to resolve inflammation and maintain immunological homeostasis in response to constant exposure of nonself molecules derived from nutrients or resident bacterial products with inflammatory potential. Accumulating evidence has shown that hepatocytes maintain local immunotolerance through direct or indirect interaction with immune cells in liver, but the underlying mechanisms remain poorly defined. Fork head box O (FoxO) transcription factors and FoxO1 in particular, are critical liver homeostasis regulators to orchestrate gene expression upon cellular stress. Here we aimed to investigate the involvement of hepatocyte FoxO1 in maintenance of immunological homeostasis. We generated hepatocyte-specific FoxO1 knockout (CKO) and control (WT) mice and compared the gene expression difference.
