Human induced pluripotent stem cell (iPSC)-derived cardiomyocytes cultured in 3-dimensional suspension culture were treated with AS1842856 FOXO-inhibitor (n=3 independent wells) or DMSO vehicle control (n=3 independent wells). Total RNA was isolated from each well and utilized for RNA sequencing. Overall design: Human iPSC-derived cardiomyocyte 3D cultures were treated with AS1842856 (1µM) FOXO-inhibitor or DMSO (0.2%) vehicle control, for 5 days starting at Day 9 of differentiation. They were maintained in RPMI 1640 media supplemented with B27 and Insulin. UCSD142i-86-1 iPSC line (female 116 donor, fibroblast-derived) was obtained from WiCell and utilized following institutional guidelines.
