Gene expression profile of WT and PD-1 -/- CD8+ T cells from murine skin (ear)

Gene expression changes were computed between naïve, WT and PD-1 -/- T Cells. WT or PD-1 -/- OT-1+ CD8+ donor T cells were sorted from the ear skin of recipient mice at day 14 after VACV-OVA skin scarification. Naïve CD8+ OT-1 T cells were isolated from lymph node and spleen of naive mice by negative selection on MACS beads Overall design: 1X10^5 WT CD8+ OT-1 T cells were mixed with 1x10^5 PD-1 -/- CD8+ OT-1 T cells and were co-adoptively transferred to WT recipients one day before VACV-OVA infection. Recipient mice were vaccinated by VACV-OVA skin scarification on each ear with 1x10^6 and on the base of the tail with 2x10^6 viral p.f.u. At day 14, WT or PD-1-/- donor CD8+ T cells were isolated from ear skin, sorted into Trizol medium and sequenced. Naive CD8+ OT-1 T cells were isolated from lymph node and spleen of naive mice by negative selection on MACS beads, stored in Trizol medium and sequenced.
