Purpose: compare the response of DRG neurons to a central axonal injury in presence of histone deacetylase inhibitor by measuring accessible chromatin and transcription Results: we found that MS-275 was able to induce changes in chromatin accessibility and gene expression. In particular, an increased accessibility correlated with changes in gene expression, and a decreased accessibility correlated with gene downregulation Method: ATACseq and RNAseq from sciatic mouse L4-L6 DRG tissue 24 hours after the following treatments: a) spinal cord dorsal column axotomy (DCA) plus 12 mg/kg MS275 intraperitoneal injection b) spinal cord dorsal column axotomy (DCA) plus vehicle (3.2% Tween80, 20% PEG300, 4% DMSO) intraperitoneal injection. MS275 or vehicle were injected intraperitoneally at the time of the injury and 2 h before sacrifice. Pool of sciatic DRGs from 1 mouse was used for each ATACseq sample. Pool of sciatic DRGs from 2 mice were used for each RNAseq sample.DRG were collected and used fresh for ATAC-seq. DRG were collected in RNase later reagent, and crushed with RNase free micropestle for RNA extraction. Biological triplicates of each condition were generated.
