资源简介:
Sodium Hypochlorite has shown improvements to wound healing in both animal and human studies. This data was generated as part of investigating the potential mechanism behind this action.
First, the MIC and MBC of this hypochlorite in saline formulation was evaluated, and found to be 7mmol/L unilaterally eliminated the species tested. This informed further work detailed below
In the presence of HIFBS, a suitable organic load to mimic topical application of hypochlorite onto wounds, IL-6 or IL-10 were dialysed against hypochlorite of varying concentrations in isotonic saline. This was then dialysed against saline alone for a minimum of 30 minutes.
human IL-6 ELISAs were then used to identify the rate and severity of IL-6 degradation by this formulation. This was followed by the same samples being analysed utilising Hek-Blue reporter cell lines to test for cytokine function.
It was found that, at the concentration capable of eliminating all bacteria, hypochlorite has a differential effect upon IL-6 and IL-10, both structurally (ELISA) and functionally (Bioassay).
in a wound this would lead to an anti-inflammatory effect, potentially explaining the wound healing improvements seen in other publications.
Lastly, the cytotoxicity of this formulation was tested against the skin fibroblast cell line HFF-1, to investigate the tolerability profile.
