Biological mechanisms explaining asthma control improvement following bronchial thermoplasty are still not well understood. We used a large-scale transcriptomic approach to evaluate the transcriptome of bronchial epithelial cell (BEC) obtained before and after bronchial thermoplasty treatment (BT). We compared gene expression profile from six bronchial epithelial cell obtained before and after bronchial thermoplasty treatment in severe asthma. Total RNA was extracted using the RNeasy mini kit (Qiagen) following the supplier’s recommendations. RNA concentration was measured using a Nanodrop and their integrity by using the Agilent 2100 Bioanalyzer (Agilent Technologies). Microarray analysis was performed on 500ng RNA samples with Affymetrix human ClariomTM S assay containing 280,809 probes (Affymetrix, Santa Clara, CA) at the McGill University and Genome Quebec Innovation Centre. RNA processing steps (RNA extraction, cDNA synthesis, cDNA fragmentation and labeling, and chip hybridization) were performed in parallel for each pair of epithelial samples to minimize technical variability. The raw image files (CEL format) generated from the analysis of the scanned image were used for the statistical analysis
