Plasmodium falciparum (PfAMA1/PfMSP1)-specific heavy chain BCR sequence

Single PfAMA1/PfMSP1-probe-positive CD19+ CD10- B cells were index-sorted for CD21 and CD27 from adult Malian PBMCs. The heavy chain of the BCR was amplified using reverse transcription and PCR followed by Sanger sequencing as previously described (Andrews et al., Immunity, 2019; Tiller et al., J. Immunol. Methods. 2008). A mix of sequencing primers 3'IgGint GTT CGG GGA AGT AGT CCT TGA C and 3'IgMint GGA ATT CTC ACA GGA GAC GA were used. For complete method see Hopp et al., JEM, 2021 (doi 10.1084/jem.20200901).Using index data obtained during the flow sort, BCR sequences were then identified as CD21- CD27- atBCs, CD21- CD27+ actBCs and CD21+ CD27+ MBCs.
