We investigated the role of cardiomyocyte MuRF1 in the transcriptional responses to cyclic stretch (15% biaxial stretch at 1 Hz for 60 min) using the HL-1 cell line. We transiently knocked-down MuRF1 expression in HL-1 cells in 6-well BioFlex® culture plates using Ad.shRNA MuRF1 (or scramble control) transduction for 48 hours (resulting in >50% protein knock-down). Cells were then stretched at 15% uniaxial stretch at 1 Hz for 60 min in a computer-regulated Flexcell® FX-5000™ Compression System (Flexcell International Corporation, Hillsborough, NC), with parallel non-stretched cells (0 min stretch) used as a control.
