Adar +/- vs Adar Za/- primary lung fibroblasts

Primary lung fibroblasts (LFs) were derived from Adar +/- x Mavs-/- and ADAR1 Za/- x Mavs-/- (Za domain mutations: N175A/Y179A) and left untreated or stimulated with 200 U/ml of hybrid IFNa-B/D for 16 hours. For each group, the LFs were derived from three individual mice. In addition, 2 biological replicates of IFNa-B/D-treated ADAR1 -/- x Mavs-/- were included in this study. RNA extraction was performed using RNeasy columns (QIAGEN) with on-column DNase I digestion. RNA integrity was tested with the Agilent RNA 6000 Pico Kit. Sequencing libraries were prepared using the Stranded mRNA ligation kit (Illumina) with the Ribo-Zero Gold kit (Illumina) for ribosomal depletion. Deep sequencing of the libraries was performed on an Illumina Novaseq6000 generating 150 bp paired-end sequencing reads.
