Validation of 21 differentially methylated regions in childhood pre-B cell acute lymphoblastic leukemia using amplicon bisulfite sequencing.

A large number of differentially methylated regions (DMRs) with a similar distribution pattern across different genomic elements were discovered by methylome analysis of five pairs of twins . The DMRs located in coding genes and known regulatory elements were identified and examined in a further cohort of 150 sibling pair acute lymphoblastic leukemia (ALL) patients. We validated SNPs and indels in these coding regions and upstream regulatory region of genes that potentially have functional effects. Additionally we applied high-throughput MiSeq amplicon sequencing on libraries constructed by a bisulfite sequencing PCR (BSP)-based procedure, we successfully amplified and sequenced 21 DMRs. The data presented here represent the 150 ALL sibling pairs only, the data used in the original investigation of the five pairs of twins is available from the SRA and GEO databases under the accesions SRP045691 and GSE59869 respectively.
