We used 3 established GBM cell lines to generate TTFields-resistant cells using Inovitro system. Overall design: To generate TTFields-resistant GBM cells, we utilized the Inovitro™ system to treat GBM cells continuously with TTFields (200 kHz unless otherwise noted) until resistant cells emerged. To ensure that changes in proliferative rates were not due to culture confluency and to determine temporal changes in gene expression as resistance developed, we maintained a constant growth condition by reseeding cells at the same starting density every seven days (7D) of TTFields treatment followed by a two-day treatment break to allow cells to recover before a new treatment cycle resumed. Numbers of live cells were enumerated on days two, four, and seven of each treatment cycle. At different indicating time points, total RNA was also collected for RNA-sequencing (RNA-seq) from the remaining cells. A maximum of five treatment cycles were performed for each cell line. For control, the same cells were subjected to the identical number of culturing cycles without TTFields treatment (non-TTFields-treated or NT). The libraries are named as {sample_name}_{batch}_{replicate_number}_{technical_replicate}. There is only one library that has a techincal replicate. While analyzing the data, make sure to correct for any batch effect as recorded in the "batch" field.
