<b>Raw data related to the paper entitled:</b> Optical control of spatially localized red blood cell activity by holographic tweezing<b>Data type:</b> Microscopy images<b>Imaging modality:</b> High-speed brightfield microscopy<b>Camera:</b> PROMON U750 high-speed CMOS camera (AOS Technologies)<b>Microscope:</b> Custom-built microscope<b>Objective:</b> Ob2 100x achromat oil-immersion objective with NA=1.45 (Nikon)<b>Tube lens:</b> 20 cm focal length<b>Sample:</b> Red blood cells (RBCs) in phosphate-buffered saline (PBS) deposited on glass<b>Data description:</b>The dataset consists of TIF images of RBCs captured using a high-speed CMOS camera operating at 1000 fps. The images were acquired using a custom-built microscope with a 100x oil-immersion objective and a 20 cm focal length tube lens. The RBCs were suspended in PBS and deposited on a glass coverslip.The dataset includes two types of images:<b>Free-cell images:</b> These images show RBCs freely flowing in PBS.<b>Trapped-cell images:</b> These images show RBCs trapped by holographic optical tweezers (HOT).The HOT are generated by a spatial light modulator (SLM) and focused onto the sample using a microscope objective. The SLM is programmed to create a pattern of light that traps the RBCs at specific locations.The dataset can be used to study the effects of optical trapping on RBCs. The data can also be used to develop and test new methods for optical manipulation of RBCs.<b>Data organization:</b>The dataset is organized into two compressed files .rar:<b>Free_cells:</b> This contains the free-cell images.<b>Trapped_cells:</b> This contains the trapped-cell images.Each folder contains a set of subfolder that specify the cel number. Each subfolder contains 5000 TIF files. The filenames are in the format <code>captura_p0001.tif </code>for example, for the first frame of the image sequence.
