Effect of Tubeimoside I (TA) on the innate immunity in C57BL/6 mice

A saponin monomer Tubeimoside I (TA) was found to have potential adjuvant activity. Since the adaptive immune response is mediated by innate immunity, we took the immune injection site (quadriceps femoris muscle) as the local immune microenvironment, and injected TA (50 μg) into quadriceps muscle to prepare samples. Muscles injected with PBS were served as the negative control. RNA was extracted 4 hours after drug treatment, and samples were prepared for whole-gene transcriptome expression profile microarray detection. Mice were intramuscularly injected in the quadriceps with 50 μg TA dissolved in 50 µL of PBS, with each mouse injected in one leg only. The corresponding contralateral quadriceps were injected with an equal volume of PBS serving as the negative control. Quadriceps muscles were collected at 4 h after injection. The total RNA was extracted for microarray detection.
