Oak (Quercus robur 3P genotype) BAC sequences. Oak BAC sequences

Two BAC libraries were constructed for Quercus robur genotype 3P. These libraries were constructed in two different cloning sites (EcoRI, HindIII), in order to enhance unbiased genome representation which are due to uneven distribution of restriction sites. Firstly, the BAC libraries were fully characterized in depth by analyzing average insert size, degree of organelle contamination, empty wells. Additionally, three sets of BAC clones were screened and sequenced i/allelic BACs, to validate the assembly procedure of the diploid genome sequence, i.e. the presence of distincts scaffolds (haplotypes) corresponding to their allelic BAC counterparts, ii/ BACs carrying expressional or positional candidate genes coinciding with QTLs for water-use efficiency, bud burst or epinasty, for further studies aiming at characterizing QTL regions of adaptive traits, and iii/ BACs taken at random or using BAC ends.
