Total starch content is expressed as percentage of fresh weight (water content in flours was about 9%, with no significant differences among samples, see Table S1 in File S1). Amylose was detected either by enzymatic or colorimetric methods and it is expressed as a percentage of total starch. To facilitate comparisons, all values are also reported in brackets as percentage of the corresponding wild type value. Total starch and relative amylose content was determined on two biological replicates. Significant differences were detected by Student's t-test (P<0.05 = *; P<0.01 = **). For comparison, colorimetric analysis of amylose was performed on a single biological sample for each genotype, and data shown are means of 3 technical replicates (standard deviations were in all cases below 10% of the mean value).
