RAW DATA LC-MS of TB-PMN

The peptide samples were suspended in 10 µl of 0.1% formic acid and transferred into low-protein binding tubes. The samples were centrifuged at 8000 g for 10 min and transferred into new tubes. Then, 4.5 µl of each peptide sample was injected into a LC-MS/MS analyzer (Hybrid quadrupole Q-TOF impact II™, Bruker Daltonics, United States). Separation of tryptic peptides was performed using Ultimate3000 Nano/Capillary LC System (Thermo Scientific, UK) coupled with a Nano-captive spray ion source. Mobile phase A (0.1% formic acid) was added into the column to transfer the samples with a flow rate of 15 µl/min. Mobile phase B (5–50% solution of 0.1% formic acid in 80% acetonitrile) was added for peptide separation with a flow rate of 600 nl/ min for 15 min. Electrospray ionization was carried out at 1.6 kV using the CaptiveSpray. Then, the column was rinsed thrice with 80% mobile phase B at 35°C. The MS/MS survey was set to separate proteins ranging from 150 to 3,000 Da with 0.5 sec scan time.
