This study investigates the transcriptomic mechanisms following intervention with Panax quinquefolius-Notoginseng-Salvia miltiorrhiza granules (XYS granules) in a rat model of vascular dementia. Vascular dementia was induced in male Sprague-Dawley rats by permanent bilateral common carotid artery occlusion (BCCAO). Animals were randomly divided into three groups: sham-operated control, BCCAO model, and BCCAO + XYS granules. After 4 months of drug administration, hippocampal and cortical tissues were harvested for total RNA extraction. Strand-specific paired-end RNA sequencing was performed on the Illumina NovaSeq 6000 platform. Raw sequencing data were processed for quality control, alignment to the Rattus norvegicus reference genome (Rnor_6.0), and quantification of gene expression levels. Differential expression analysis was conducted to identify genes and pathways associated with neuroinflammation, synaptic plasticity, and cognitive function. This dataset provides comprehensive transcriptomic resources for understanding the molecular basis of herbal medicine intervention in vascular dementia and may facilitate the discovery of novel therapeutic targets.
