Enhancers, characterized by H3K4me1 and H3K27ac histone marks, are critical determinants of cell identity and drive lineage-specific gene expression patterns. Erythropoietin (Epo) is the hormone that initiates terminal erythroid differentiation, and thus may function in part to modulate the epigenome of erythroid cells. Although enhancers relevant to erythroid cells have been identified, the mechanism by which Epo alters the dynamics of histone modifications has not been defined. Here, we identified and classified the repertoire of enhancers present in proerythroblasts following Epo stimulation using ChIP-exo analysis for H3K4me1, H3K27ac, and H3K4me3 histone marks and Tal1 occupancy in a model murine system.
