Research objective: Development of a live-cell, single-ribosome-resolution assay (stopless-ORF circular RNAs; “socRNAs”) to precisely measure translation elongation kinetics and ribosome–ribosome interactions in cells, and use it to test how ribosome collisions affect pausing, recycling, and overall translation efficiencyType of research: Quantitative fluorescence imaging (single-molecule imaging in living cells)Method of data collection: Live-cell fluorescence imaging of U2OS cells transfected with socRNA reporters. socRNAs tethered to the plasma membrane to aid tracking, and translation was visualized using the SunTag system. Spot intensities were measured over time under different conditions (different reporters, or different different drugs). Type of data: Live-cell imaging data acquired with a spinning disc confocal microscope. Data is provided as tif files and can be opened with conventional image analysis tools (e.g. Fiji).
