To find epigenetic factors potentially involved in Sorafenib resistance in hepatocellular carcinoma cells, an epigenetic factors-targeted CRISPR/Cas9 library was applied to perform the screening. The CRISPR/Cas9 library which contains 5,648 sgRNAs (cloned into the pLentiCRISPRv2 vector) targeting epigenetic factors (containing 131 non-targeting controls) was packaged into lentivirus particles and used to infect HepG2 cells to generate a cell pool. The cell pool was treated with DMSO or 5 μM Sorafenib for 15 days, or not treated. Then, the fragments containing sgRNAs were amplified by PCR, and subjected to high-throughput sequencing.
