Mice transplanted with BRSST (Bcor, Runx1, Sh2b3, Stag2, Tet2) sgRNAs in addition to the sgEzh2 develop myeloid leukemia with a significantly earlier onset compared to missing sgLuc. We used global expression analysis by microarray to determine regulated gene programs after addition of the Ezh2 sgRNA. Overall design: LSK cells expressing SpCas9 were transduced with the sgRNA combination and transplanted into irradiated C57Bl/6J mice. BM was harvested after development of a leukemia or after 6 month and used for RNA extraction.
