Single nucleus RNA-seq atlas of mouse Lepr-expressing hypothalamic cells

To uncover the cellular identifies of the mouse hypothalamic Lepr cells, we used single nucleus RNA-seq (snRNA-seq) from enriched hypothalamic Lepr cells. Lepr-Cre;R26-LSL-Sun1-sfGFP mice were maintained on ad libitum chow prior to microdissection of the hypothalamus. We then isolated nuclei, FACS sorted for GFP-positive nuclei, and subjected these nuclei to snRNA-seq using the 10x Genomics 3' gene expression platform.
为解析小鼠下丘脑瘦素受体(Lepr)阳性细胞的细胞身份,我们以富集的下丘脑Lepr阳性细胞为研究材料开展单细胞核RNA测序(single nucleus RNA-seq,snRNA-seq)。实验中所用的Lepr-Cre;R26-LSL-Sun1-sfGFP转基因小鼠,在进行下丘脑显微解剖前均以自由进食普通饲料的方式饲养。随后我们分离细胞核,通过荧光激活细胞分选(Fluorescence-Activated Cell Sorting,FACS)筛选GFP阳性细胞核,并利用10x Genomics 3'端基因表达平台对这些细胞核完成单细胞核RNA测序。
