We set out to systematically decipher the OCT4-RNA interactome by HITS-CLIP. Overall design: We utilized a CRISPR/Cas9-edited H1 cell line in which a TAP (Tandem Affinity Purification) tag was knocked in to the endogenous POU5F1 locus to facilitate the isolation and enrichment of OCT4-RNA complexes.
