We performed single cell transcriptome analysis of Treg cells (CD4+CD25+) from healthy Mycfl/flFoxp3Cre/WT female mice, in which Myc-sufficient (WT) and Myc-deficient (KO) Treg cells co-exist due to random inactivation of the X chromosome that harbors the Foxp3 gene. Overall design: Cells (CD4+ CD25+ YFP+ and YFP- cells from Mycfl/flFoxp3Cre/WT mice) were sorted by flow cytometry into a well of a chilled 96-well plate (filled with 14 µl PBS with 0.06% BSA). Cells were encapsulated in one lane of a 10x Chromium single cell controller. Single- cell transcriptome library is prepared with a Single Cell 3' Reagent Kit (V2 chemistry) following standard protocol
