To determine the gene expression changes in the retina of 2-month-old Rd10 mice reared in darkness throughout their lifespan, we collected retina samples from Rd10 and WT (C57BL/6J mice) mice, generated single-cell suspensions, and performed scRNA-Seq (10X Genomics). One group of Rd10 mice was treated for 1 month via dietary administration of tamsulosin, metoprolol and bromocriptine drug cocktail. Vehicle- and TMB-treated Rd10 mice: One retina from four female Rd10 mice were pooled to prepare single-cell suspensions at a final concentration of 1,000 cells/µl. WT mice: One retina from two female and two male C57BL/6J mice were pooled to prepare single-cell suspensions at a final concentration of 1,000 cells/µl. For each group, freshly dissociated cells (~16,500) were loaded into a 10x Genomics Chromium Single Cell system using v3 chemistry following the manufacturer's instructions.
