In this study, by using 11-12 months old embryos, makapuno plants are successfully produced after 5-6 months of in vitro culture. Cutting of primary root which produced more new roots is a key factor for rapid plant growth and for ensuring high survival rate at the hardening stage. This is the first report on root cutting of makapuno embryo culture and its related gene expression study. We studied the occurrence and expression of genes related to root cutting and new root development which could be useful for the future study of molecular mechanism in palm root development. Though many works on the improvement of makapuno embryo rescue have been reported, our protocol is simple, fast and efficient, which can be easily applied for the commercial production of this valuable coconut plants. Consequently, this would have an impact on the way of improving income of coconut growers.
