The effect of AEBP1 and PELP1 deletion was studied in human aortic endothelial cells exposed to atheroprone WSS, using the orbital system. Overall design: Human aortic endothelial cells (HAoECs) were seeded onto gelatin-coated six-well plates and transfected with AEBP1 siRNA (siAEBP1), PELP1 siRNA (siPELP1), or scrambled control siRNA (sictrl) using the Lipofectamine RNAiMAX transfection system. After a six-hour incubation, the media was replaced, and cells were exposed to flow on an orbital shaker for 72 hours. Cells subjected to disturbed flow in the center of the well were scratched, and total RNA was extracted and analyzed via bulk RNA sequencing.
