This study was carried out to compare changes in gene expression in 120 hpf livers that overexpress as3mt in hepatocytes between after 24-hour exposure to 1.0 and 1.5 mM iAs. Tg(fabp10a:As3mt) untreated = CL 1 AS3MT 0; Tg(fabp10a:As3mt) iAs treated = CL 1 AS3MT 1 or CL 1 AS3MT 1.5. Overall design: The samples in this series include only livers from 120 hpf zebrafish larvae that over express as3mt in hepatocytes with and without exposure to 1 and 1.5 mM of inoragnic arsenic (iAs) from 96 - 120 hpf. This study was carried out by comparing RNA extracted from 120 hpf zebrafish livers from six groups: untreated or 24-hour exposure to 1 mM or 1.5 mM iAs in CAAX-EGFP positive larvae and CAAX-EGFP positive and as3mt overexpressing larvae. To generate these samples, (tg:(fab10a:as3mt, cry:dsRed) were outcrossed to tg(fabp10a:CAAX-EGFP) and the larvae that only had the CAAX-EGFP expression in the liver and cry:dsRed in the eye were collected. RNA from 12 livers per group collected from 5 clutches (biological replicates) per group were extracted using Trizol/Chlorofom, PolyA selected and sequenced on Illumina NextSeq550.
