RNAseq analysis of human neutrophils from healthy controls and from people with systemic lupus eruthematosus

We report gene expression profiles of blood neutrophils from (1) healthy adults (2) healthy pediatrics, (3) adults with SLE, (4) pediatrics with SLE. Participants were recruited from two sites, Liverpool UK or Bangkok, Thailand. Overall design: Neutrophils (purity >97%) were isolated from peripheral blood by separation of leukocytes from erythrocytes using HetaSep, then centrifugation through density-gradient reagent (HetaSep). Remaining erythrocytes were removed by hypotonic lysis using ammonium chloride buffer. RNA was isolated using Trizol:chloroform, cleaned up by Qiagen RNeasy kit including on-column DNase digest and sequenced on the DNBseq platform.
