Identification of RNAs regulated by ZMAT3 and p53

Identify the transcriptome regulated by p53 and ZMAT3 in HCT116 colorectal cancer cell line. The hypothsis of this study is that ZMAT3 regulates the expression of a subset of RNAs that are also regulated by p53 to mediate its biological functions. Overall design: To identify the RNAs regulated by p53, mRNA-seq was performed from total RNA isolated from HCT116 cells transfected with a control (CTRL) siRNA or p53 siRNAs (SMART pool of 4 siRNAs) in biological duplicates for 72 hours. To identify the RNAs regulated by ZMAT3, mRNA-seq was performed from total RNA isolated from a ZMAT3-wild type clone and a ZMAT3 knockout clone in biological triplicates.
