Genome-wide Cross-trait Analysis Reveals Shared Genetic Architecture between Various Cancers and Myasthenia Gravis

Myasthenia Gravis (MG) and various cancers may share common genetic foundations. This study employed large-scale genetic data to investigate their shared genetic architecture by analyzing genome-wide association study data from nine cancer types and three MG subtypes. Linkage disequilibrium score regression , pleiotropy analysis , and Mendelian randomization were applied. Results revealed significant genetic correlations in six cancer-MG pairs, particularly between renal cancer and MG. We identified 162 pleiotropic SNPs corresponding to 30 independent risk loci, with functional enrichment analysis indicating specific expression of these genes in lymphoid tissues.MR analysis demonstrated that early-onset MG was negatively associated with renal cancer risk, whereas late-onset MG was positively associated with breast cancer risk. These findings reveal a shared genetic architecture between cancers and MG, providing new insights into common mechanisms and potential therapeutic strategies.
