Gut microbiome to treat obesity in mice

Gut microbiome is associated with various diseases including cancer, metabolic syndrome and obesity. Previous studies reported that administration of Lactobacillus spp. ameliorates diet-induced obesity on clinical and preclinical studies. However, underlying modulatory functions of Lactobacillus spp. have not been validated. Here, we demonstrated anti-obesity mechanisms of Lactobacillus spp. through transcriptome analysis of bacteria and mouse gonadal white adipose tissue, metabolome analysis of bacterial culture supernatant and mice serum. In HFD mice model, administration of Lactobacillus fermentum LM1016 ameliorates diet-induced obesity, insulin resistance and fatty liver. Transcriptome analysis showed that administration of L. fermentum LM1016 inhibits inflammation as well as improve oxidative stress, and metabolomic profiling suggested that linoleic acid from bacterial culture supernatant inhibits diet-induced obesity. Our data indicate that L. fermentum LM1016 can be applied to humans as a therapeutic tool for the obesity and metabolic syndrome.
