This dataset contains transcriptomic profiles of immortalized human keratinocytes (HaCaT) cultured under high glucose conditions following ETV4 knockdown. RNA sequencing was performed using the Illumina platform to investigate how ETV4 suppression modulates gene expression in a diabetic-like microenvironment. The experimental design compared two groups: high glucose control (HG) and ETV4-knockdown under high glucose (HG+siETV4). Bioinformatics analysis revealed differentially expressed genes. Pathway enrichment analysis demonstrated significant alterations in inflammatory response, extracellular matrix organization, and apoptosis pathways. Transcription factor binding analysis confirmed enrichment of ETS family motifs among regulated genes, supporting ETV4's role as a transcriptional hub in hyperglycemic stress. These findings provide mechanistic insights into diabetic skin complications and identify ETV4 as a potential therapeutic target. The dataset enables further exploration of ETV4-regulated networks and biomarker discovery for diabetic dermatopathies.
