Healdette: An Ancestry-Aware, Ethics-Transparent Antibody Generation Pipeline Healdette is a fully operational antibody and nanobody design system that integrates real protein generation models with transparent, ethical, and ancestry-informed decision-making. This pipeline enables safe, culturally-sensitive therapeutic discovery by combining: Transformer-based sequence generation using ProtGPT2 and ProtBERT Biophysical stability scoring and predicted affinity metrics Ethical filtering with full rejection tracelines Personalization based on HLA type, metabolic rate, and prior pathogen exposure Cultural awareness, incorporating Indigenous and Irish genetic ancestry in binder selection logic Unlike traditional AI tools, Healdette is not a black box. Every generated sequence includes: Metadata explaining its origin and ethical pathway Reasons for rejection if filtered out Scoring based on biological, cultural, and immunological factors All code is real. All outputs are traceable. This project was built to demonstrate how AI can be used not just to accelerate medicine, but to do so responsibly, equitably, and ethically.
