Single cell transcriptomics of the cerebral cortex in mice lacking the PRC2 gene Eed

The Polycomb Repressive Complex 2 (PRC2) regulates corticogenesis, yet how PRC2 influences cell identity in the mature brain is poorly defined. Using a mouse model in which the PRC2 gene Eed is conditionally deleted from the developing mouse dorsal telencephalon, we performed single nuclei RNA sequencing on the cortical plate of adult heterozygote and homozygote Eed knockout mice compared to controls. Using a mouse model in which Eed is conditionally knocked out from all cells derived from the dorsal telencephalon (Emx1-iCre), we performed single nuclei RNA sequencing (snRNA-seq) on the cortical plate of a control (CTRL), a heterozygous (Eed-cHet) knockout, and a homozygous (Eed-cKO) mouse.
多梳抑制复合体2(Polycomb Repressive Complex 2, PRC2)可调控皮质发生,但目前对于PRC2如何影响成熟大脑内的细胞身份仍缺乏明确认知。本研究利用一款在发育小鼠背侧端脑内条件性敲除PRC2基因Eed的小鼠模型,对成年杂合子、纯合子Eed敲除小鼠及对照小鼠的皮质板开展单细胞核RNA测序(single nuclei RNA sequencing)。本研究同时采用另一款小鼠模型:通过Emx1-iCre介导,将源自背侧端脑的所有细胞中的Eed基因条件性敲除,并对对照(CTRL)、杂合敲除(Eed-cHet)及纯合敲除(Eed-cKO)小鼠的皮质板开展单细胞核RNA测序(snRNA-seq)。
