The goal is To examine the effects of four cytokines, HGF, IL-17A, TNF-a, and IL-22, on keratinocytes and compare them to the transcriptomes of HS skin. To examine the expression and localization of HGF, lesional skin samples from patients with HS were investigated by immunohistochemical staining and single cell data analysis. Transcriptome analysis of RNA from keratinocytes stimulated with each cytokine was performed and compared to those of HS lesions. Primary normal human adult epidermal keratinocytes were purchased and cultured using EpiLife medium with Growth Supplements and Antibiotic-Antimycotic Solution . When the cells reached 80% confluence, they were trypsinized and resuspended in the medium at 2 ×105cells/ml, and 1 ml was added to each well of the 12-well plates. After overnight starvation, the cells were stimulated with recombinant human IL-17A (200 ng/ml), IL-22 (50 ng/ml), TNF-a (10 ng/ml), and HGF (100 ng/ml) for 24 hours.
