Pregnancy is associated with metabolic changes to accommodate the mother and her fetus. The gut microbiome has been shown to modulate host endogenous and exogenous metabolism. The objective of this study was to identify how the absence of the gut microbiome impacted metabolomic changes during pregnancy. We used 4 groups of C57BL/6 mice, namely conventional non-pregnant (CVNP), conventional pregnant (CVP), germ-free non-pregnant (GFNP), and germ-free pregnant (GFP), between ages 8-13 weeks. Fecal pellets were collected from the same CV mouse at baseline and on gestation day 15 (gd 15) to characterize the gut microbiome shift during pregnancy. We performed RNA-seq analysis on liver tissues to describe hepatic transcriptome and untargeted plasma metabolome via LC-MS/MS to describe metabolite changes as results of both pregnancy and the gut microbiome.
