The cDNAs of AtMYB48 and AtbHLH68 were fused with the regulatory region of the human estrogen receptor at the C terminus, and ligated between the CaMV 35S promoter and the nopline synthase terminator in pER8. The estrogen induce MYB48 and bHLH68 expression construct were transfected into Arabidopsis stem. Arabidopsis stems after bolting 15 days were treated with 10 mmol/L estradiol for 2 h, 4 h, 6 h and 8 h, respectively, and DMSO treatment was used as control. After treatments, the stems were harvested and used for total RNA isolation and RT-qPCR and transcriptional analyses.Estrogen induced expression transgenic MYB48 and bHLH68 materials were selected after estrogen induction, and total RNA was extracted for transcriptome sequencing.
