A cell-free transcription-translation (TXTL) system was used to rapidly characterize the protospacer-adjacent motif (PAM) requirements of diverse Cas12a (Cpf1) proteins. We simultaneously expressed the Cas protein and its crRNA in a solution containing a PAM library and monitored cleavage of the library using high-throughput sequencing.
