RNA-seq analysis were applied to elucidate the transcriptional differences of PHF8 wild-type and PHF8 knockout TRAMP mouse. A total of 2,092 differentially expressed genes (Fold Change > 2, or Fold Change<0.5; FDR < 0.05) with 623 down- and 1469 up-regulated genes were identifed in Phf8-KO TRAMP mice. Total RNA was extracted from tumor tissues derived from either Phf8-KO TRAMP mice or the control TRAMP mice at week 37 using Trizol reagent (Invitrogen). And RNA-Seq was performed by Shanghai NovelBio Bio-Pharm Technology Co., Ltd (Shanghai, China). A pooled RNAs from 3 mice were mixed with an equal mass of RNA to minimize variation across samples. RNA-Seq reads were filtered and mapped to Mouse genome (GRCm38, NCBI) utilizing HISAT2. HTSeq was used to calculate the gene count. Differentially expressed genes analysis was applied utilizing DESeq2 under following criteria: Fold Change > 2, or Fold Change<0.5; FDR < 0.05.
