Histone methyltransferase G9a governs the mutant Kras-driven pancreatic carcinogenesis

ATAC-seq of mouse pancreas organoids (K,KC,KC+UNC) Overall design: Pancreatic organoid cells from the LSL-KrasG12D/+ (K) , Ptf1aCre/; KrasG12D (KC) mice were examined by ATAC-seq. Organoids were cultured either with fresh media or with 5µM UNC0638 for 24hours prior to experiments. Harvested organoids were gently digested into single cell suspension using TrypLE and DNase1, and 50,000 cells were processed for ATAC-seq. Sequence reads were trimmed and aligned to the mouse genome (mm10) using BWA. For visualization, ATAC-seq signals were normalized to RPKM and smoothed by 150bp sliding windows with 20bp bin size using bamCoverage.
