Age and gender-matched wt mice and Il17d deficient mice were sensitized with 10 ug HDM on day 0, followed by five times of rechallenging from day 7 to 11, and then the lung tissues were harvested on Day14 and processed into single cell suspension. Alveolar macrophages were sorted as live single cell CD45+ CD11Chi SiglecFhi. And total RNA was extracted by using TRIzol reagent and sent for RNA sequencing.
