Stress resilience-enhancing drugs preserve tissue structure and function in degenerating retina via selective phosphodiesterase inhibition

To determine the gene expression changes in the retina of Abca4-/-Rdh8-/- (dKO) mice, we collected retina samples from dKO animals under various experimental conditions, generated single-cell suspensions, and performed scRNA-Seq (10X Genomics). Overall design: In each of the five experimental groups, four retinae from two mice were pooled to prepare single-cell suspensions at a final concentration of 1,000 cells/µl. For each group, freshly dissociated cells (~15,000) were loaded into a 10x Genomics Chromium Single Cell system using v3 chemistry following the manufacturer's instructions.
