DNA replication initiates at distinct sites in eukaryotic genomes that are called origins of DNA replications, but the genomic features that define one eukaryotic origin are not well understood. In parallel, the total number and high-resolution position of DNA replication origins in Trypanosoma brucei remains unidentified until present days. Short nascent strands (SNS) quantification is a high-resolution technique widely used for mapping of DNA replication origins. We have performed high-throughput sequencing with preserved directionality of the SNS and mapped the initiation sites (IS) of DNA replication origins in procyclic (PCF) and bloodstream (BSF) life form of the Trypanosoma brucei. The sequencing libraries are a second-strand stranded libraries (plus strand = F1R2, minus strand = F2R1).
