Mycobacterium tuberculosis Genome sequencing and assembly

The aim of this study was to identify on-target inhibitors of Mycobacterial Membrane Protein Large 3 (MmpL3) , a member of Resistance Nodulation and Division Transporters of Mycobacterial tuberculosis from GSK Tres Cantos Anti-Mycobacterial Set. The cell-based assay identified a guanidine CCI7967 to target MmpL3. CCI7967-resistant mutants harbored mutations that mapped to MmpL3, futher confirming it's on target activity. The cell-based assay developed for this study could be used to identify new MmpL3-specific scaffolds and rapidly screen for optimized analogues to facilitate their development as potent anti-tuberculars.
