In this study, we examined the impact of copper diethyldithiocarbamate (Cu(DDC)2), a metabolite of the FDA-approved drug disulfiram, on thyroid cancer cells. Mechanistically, Cu(DDC)2 targets valosin containing protein (VCP) â a key regulator of proteostasis â as well as inducing potent transcriptional responses. Overall design: Thyroid cancer 8505C cells were treated with Cu(DDC)2 (250 nM, 24 hours) versus vehicle (DMSO). RNA-Seq libraries were prepared at the Next Generation Sequencing Facility (Genomics Birmingham Genomics Service, University of Birmingham).Sequencing libraries prepared with the TruSeq Stranded Total RNA kit (Illumina Inc), from 1ug total RNA. Alignment of raw sequence reads to the human transcriptome (hg38) was performed via Rsubread and transcript abundance estimates were normalized and differentially expressed genes (DEGs) identified using a standard edgeR pipeline.
