An new in vitro approach on dedicated biochip to quantitatively and simultaneously measure the capacity of four DNA repair pathways that support DNA double Strand Breaks. This multiplex enzyme assay characterizes DSB repair pathways in lysates prepared from untreated and treated (DNA repair inhibitiors and/or genotoxic agent) cell lines, to assess basal activities and responses to drugs, respectively. The technology quantifies the incorporation of different markers on two dedicated plasmids immobilized on biochip. The combination of which reflects the activity of 4 DSB repair pathways. The total fluorescence intensity (FI) of 4 technical replicates was quantified and the mean of these 4 replicated was calculated for each pathway. Results are expressed as Fluorescence intensity. The mean of three biological replicates was also calculated. <br>
