We developed a physiologically relevant co-culture model of patient-derived glioblastoma lines with cortical-like neural spheroids to study tumor infiltration. High-content imaging confirmed directional migration along axons and spheroid invasion, suppressed by FAK and CXCR4 inhibitors. Bulk RNA sequencing identified gene expression signatures linked to invasive behavior and drug sensitivity, providing a platform to uncover molecular drivers of glioblastoma infiltration and therapeutic vulnerabilities.
