The genomes of Salmonella enterica subsp. VII str. RKS3013 were sequenced in this study. The strains were inoculated in LB agar plates and incubated at 37 degrees Celsius overnight, and single colonies were inoculated in LB medium, shaking at 37 degrees Celsius and 200rpm for 8 hours. For each strain, 4ul of the culture was transferred to 200ml fresh LB medium and grown at 37 degrees Celsius and 200rpm to logarithmic phase. The cells were collected and genome DNA was isolated with a standard phenol-chloroform extracting procedure. Paired-end 250-bp DNA libraries of an insert size of 350-400 bp were built with TruSeq DNA PCR-Free Library Preparation Kits, and Hiseq 2500 platform was used to sequence the genomes of the strains. PacBio RS II platform was also used to sequence the genomes, with an insert size of 10 kb. P6-C5 kits were used for preparation of the PacBio libraries. Spades (hybridSpades version) was applied to assemble the genome sequences with the hybrid short and long reads. RASTtk version 1.3.0 was used to annotate the gene composition and function.
